The information below comes from published research and documents what has been studied, not what should be done. Many of these compounds are investigational and not approved by the FDA; possession or use outside an authorized clinical trial may be illegal in your jurisdiction. This is not medical advice.

Ecnoglutide is a long-acting GLP-1 analog in clinical development for type 2 diabetes and obesity. It is not FDA approved.
Dosing Reference
Reconstitution
Add 2 mL bacteriostatic water to the 10 mg vial. Resulting concentration: 5 mg/mL.
| Dose | Syringe Units | mL Volume | Schedule |
|---|---|---|---|
| 0.6 mg | 12 units | 0.12 mL | SubQ, once weeklyPhase 3 starting dose |
| 1.2 mg | 24 units | 0.24 mL | SubQ, once weeklyPhase 3 mid dose |
| 2.4 mg | 48 units | 0.48 mL | SubQ, once weeklyPhase 3 target dose |
Math assumes U-100 insulin syringes (1 mL = 100 units). Verify your syringe matches before use. Round half-units to the nearest visible mark.
Reconstitution
Ecnoglutide (ecoglutide / IBI362 variant) is an investigational GLP-1 receptor agonist developed in China, approved there for weight management. It is supplied as a pre-filled pen or lyophilized vial for subcutaneous use. Compounded research-peptide forms are not FDA-approved.
For a compounded 10 mg vial reconstituted with 2 mL, the concentration is 5 mg/mL; a 1.2 mg dose draws to 24 units (0.24 mL) on a U-100 syringe. Store per manufacturer directions; compounded forms are not FDA-approved.
Cycling Protocol
Approved-in-China / phase 3 protocols titrate ecnoglutide weekly over several weeks to a target dose, then maintained long-term. No fixed cycle end; studied as chronic weekly therapy.
Routes of Administration
Subcutaneous injection (only established route): Once weekly into the abdomen, thigh, or upper arm. Any time of day, independent of meals.
No oral, intranasal, or intravenous route is established. Compounded forms are not FDA-approved.
Stacking Protocols
| Stack | Components | Purpose | | --- | --- | --- | | GLP-1 + Lifestyle | Ecnoglutide + diet/exercise | Approved-in-China weight protocol |
Combining multiple GLP-1 agonists is not recommended. Ecnoglutide is approved in China but not FDA-approved in the US.
Side Effects & Safety
- Nausea, vomiting, diarrhea, constipation — common, usually transient
- Hypoglycemia — risk, especially combined with insulin/sulfonylureas
- Pancreatitis — rare; discontinue if suspected
- Injection-site reactions — common
- Thyroid C-cell tumors — in rodents; avoid in MTC/MEN2 history
Ecnoglutide is approved in China but not FDA-approved in the US. Compounded research-peptide forms are not FDA-approved.
Research Basis
Ecnoglutide is an investigational/approved-in-China GLP-1 receptor agonist with an extended half-life enabling once-weekly dosing.
Mechanism: GLP-1 receptor agonism — glucose-dependent insulin secretion, glucagon suppression, slowed gastric emptying, appetite reduction.
Clinical evidence: Approved in China for weight management based on phase 3 trials demonstrating weight loss. Not FDA-approved in the US.
Approved in China; not FDA-approved in the US. Compounded research-peptide forms are not FDA-approved.
